Overview

[197Hg]Hg-gold nanoparticles are a form of radiation nanomedicine in which the Auger electron-emitting radioisotope mercury-197 (^197Hg) is incorporated into gold nanoparticles (AuNPs) via the synthesis of a mercury-gold amalgam. These nanoparticles can be used alone (non-targeted) or conjugated to targeting molecules, such as the anti-EGFR antibody panitumumab, for specific delivery to tumor cells overexpressing EGFR, notably in glioblastoma multiforme (GBM). The mechanism involves delivery of highly localized radiation to tumor cells through emission of Auger electrons from ^197Hg, causing cytotoxic DNA double-strand breaks. Delivery is achieved via convection-enhanced delivery (CED) directly into brain tumors, maximizing tumor absorption and minimizing systemic exposure. These nanoparticles are an investigational radiopharmaceutical intended primarily for the treatment of GBM, with preclinical studies demonstrating efficacy in targeted cell killing and tumor suppression[1][3][5][7].
